The big cats sneaked out of Kruger park - a major tourist attraction which is almost the size of Belgium - on Sunday night.
They have now been spotted in Matsulu village outside the park.
Livestock as well as people are at risk of attack as the area around Kruger also contains farms which raise cattle.
Rangers are now searching for the animals and residents have been warned to be "extra vigilant" and report any sightings of the creatures.
The park is a major tourist attraction in South Africa
The park, which covers an area of 7,523 sq miles and borders Zimbabwe and Mozambique, is home to about 1,500 lions.
The latest incident comes two months after five other lions escaped from the park.
Four were re-captured in neighbouring farms and one is still on the loose.
Officials have said animals sometimes sneak out through dry river beds, or use holes dug out by other creatures near barrier fences.
